Terpenoid aldehydes from cotton, such as hemigossypol, gossypol, and the heliocides, are important components of disease and herbivory resistance in cotton. Manipulation of the biosynthetic pathways for these compounds provides an opportunity to improve cotton resistance to pathogens and insects. A cytochrome P450 CYP82D109 was identified from cotton. Suppression of this gene through RNAi resulted in over 90% reduction in the total gossypol-heliocide pathway compounds but increased the induction of lacinilene C pathway compounds. The resulting RNAi plants showed increase resistance against Fusarium wilt pathogen and decreased toxicity against cotton insect pest Helicoverpa zea.

1.      Toward Control Strategies of Emerging Pathogens and Nematodes of Cotton:

Determine whether fusaric acid contributes to differences in virulence among races and biotypes of Fusarium oxysporum f. sp. vasinfectum to provide a basis for developing strategies for controlling Fusarium wilt of cotton. (2007-2012, USDA-ARS National Program)

2.      Identification and Characterization of a Dirigent Protein That Controls the Biosynthesis of (+)-Gossypol in Cotton (2010-2014, Cooperative Agreement).

3.      Development of Cotton Plants with Gossypol Free Seed and Normal Glanded Foliage (2002-2011, obtained external funding in excess of $597,000).

4.      Identification of Genes Coding for Production of Fusaric Acid in Highly Virulent Strains of Fusarium oxysporum vasinfectum.  (2009-2011, obtained external funding of $60,000).

5.       The Role of Fungal Secondary Metabolites in Pathogenicity of Wilt Pathogens Affecting Cotton. (2009-2014, Cooperative Agreement)
